Surrozen Clears FDA IND Hurdle for DME Drug, Unlocking $95.1 Million Financing Milestone

Key Points

  • The FDA has cleared Surrozen’s investigational new drug application for SZN-8141, enabling clinical development in diabetic macular edema.
  • Surrozen expects to dose the first patient in its Phase 1b/2a DUET study in the fourth quarter of 2026, with initial data anticipated in the second half of 2027.
  • The IND clearance satisfies a milestone for the expected second closing of Surrozen’s March 2025 private placement.
  • The second closing is expected to provide approximately $95.1 million in gross proceeds before fees and expenses, subject to remaining closing conditions.
  • In the Phase 2a portion of DUET, Surrozen plans to compare two SZN-8141 dose levels with Vabysmo in approximately 60 treatment-naïve DME patients.

Surrozen (NASDAQ: SRZN) has announced that the FDA has cleared its investigational new drug application for SZN-8141, allowing the company to begin clinical testing of the diabetic macular edema treatment. The IND milestone also triggers the expected second closing of Surrozen’s March 2025 private placement, which is expected to deliver approximately $95.1 million in gross proceeds.

SZN-8141 FDA IND Clearance Enables DUET Phase 1b/2a DME Trial

SZN-8141 is an investigational intravitreal injection designed for diabetic macular edema, or DME. It combines Frizzled 4-mediated Wnt agonism with VEGF antagonism, a dual approach Surrozen believes may differentiate the therapy from treatments that act through a single mechanism.

Surrozen expects to dose the first patient in the DUET Phase 1b/2a study during the fourth quarter of 2026. Initial data are expected in the second half of 2027. SZN-8141 is not approved for any indication.

DUET Trial Will Compare SZN-8141 With Vabysmo in Phase 2a

The open-label Phase 1b portion will evaluate a single ascending dose of SZN-8141 in treatment-naïve and previously treated DME patients. Participants will receive one intravitreal injection and be followed for about three months for safety, tolerability, pharmacokinetics, immunogenicity and exploratory measurements of vision and retinal anatomy.

The randomized, double-masked Phase 2a expansion is expected to compare two SZN-8141 dose levels with Vabysmo (faricimab-svoa) in approximately 60 treatment-naïve DME patients. Patients are planned to receive three monthly doses followed by a four-month follow-up period, with measures including best-corrected visual acuity, optical coherence tomography, OCT angiography and ultra-widefield fluorescein angiography.

IND Milestone Triggers Expected $95.1 Million Private Placement Closing

The effective IND satisfies the condition for the second closing of Surrozen’s March 2025 two-tranche private placement. Subject to the agreement’s remaining closing conditions, the company expects the transaction to close around October 20, 2026.

Surrozen expects gross proceeds of approximately $95.1 million before placement-agent fees and other expenses. The company plans to use the funds to advance SZN-8141, develop SZN-8143 and support broader operations. After the closing, Surrozen expects to file a Form S-3 resale registration statement for shares and warrants issued in the second closing.

Surrozen Plans Broader Retinal Disease Development

Surrozen is also developing SZN-8141 for wet age-related macular degeneration and is evaluating other retinal vascular-disease applications. Its separate preclinical candidate, SZN-8143, combines FZD4 agonism, VEGF antagonism and IL-6 antagonism and is being developed for DME, wet AMD and uveitic macular edema.

For SRZN investors, the IND clearance removes a near-term regulatory gate and supplies an expected financing catalyst. The key remaining risks are clinical execution, safety, the ability to show meaningful activity against existing therapies and completion of the private placement under its stated conditions.
